ATF SP3 is used for two components:
1) ASC (with reservoir in truck)
2) part of the rear-differential
The ASC component does NOT need to be drained or refilled (this part holds over 7 quarts I believe).
However, the rear-differential component that takes ATF fluid should be drained/refilled. That component uses less than 1 quart. You will see fill and drain bolts with arrows pointing to them with "ATF" labeled.

Yes, make sure you find the correct fill plug and can loosen it before draining. Loosening the fill plug first is always a good idea in case you have problems (that way you don't drain the fluid and make the car undriveable if the fill plug is stuck for some reason).

Not sure if you can use just any ATF. I would use mitsu ATF SP3 as specified. Just fill it until it starts to come pouring out. I usually will let it spill out for at least 10 minutes before tightening the fill plug. It's important that the car is level though.
N118GT, I think there are other bolts that are labeled to help you know if you're on the LSD gear oil side or the ATF SP3 side of the rear diff, but they are not all for filling. Look at the link I posted to see the service manual guide to where to drain/fill from.
